B vitamins have always had a reputation as the “energy” vitamins, the ones on every multivitamin label promising to fight tiredness. What’s newer is the evidence for a second role: several of them, particularly B6, appear to genuinely support how your brain handles stress, not just how tired you feel.

What Are B Vitamins, and Why Do They Matter for the Nervous System?
B vitamins are a family of eight water-soluble vitamins: thiamine (B1), riboflavin (B2), niacin (B3), pantothenic acid (B5), pyridoxine (B6), biotin (B7), folate (B9), and B12. They work as cofactors, helper molecules the body needs to run chemical reactions, rather than as fuel in their own right.
B6 has the most direct evidence for mood and stress, covered next. B12 keeps the protective coating around nerve fibres, called myelin, intact. Folate and B12 both help regulate homocysteine, an amino acid that becomes harmful to the nervous system and heart at raised levels, covered in more detail further down. Because none of the B vitamins are stored in large amounts except B12, a steady daily supply matters.
Where the Evidence Is Strongest: B6 and Mood
This is where the evidence for B6 is strongest. A double-blind randomised controlled trial published in August 2026 gave 268 adults either 100mg of daily vitamin B6, B12, or a placebo for one month. Participants with depression symptoms at the start of the trial saw a large reduction on the B6 arm compared with both other groups. Those just below the clinical threshold for mild depression saw a smaller, but real, benefit too [1].
It builds directly on the same Reading University team’s earlier trial, which gave over 300 adults a high dose of B6, roughly 50 times the standard recommended daily amount, for one month. Participants reported feeling less anxious and less low in mood, an effect the researchers described as small compared with medication but statistically reliable [2]. Follow-up testing found the effect tracked with a genuine rise in GABA activity in the brain, the same calming neurotransmitter targeted by anti-anxiety medication. A rise in serotonin production, the neurotransmitter most linked to mood regulation, was found too [2].
Neither of these trials stands alone. An earlier randomised trial gave 60 office workers a high-dose B-complex supplement for 90 days and found significantly lower personal strain and reduced confusion and low mood by the 12-week mark, compared with placebo [3]. Three separate trials, three different research groups, all pointing the same way is a much stronger signal than any one study alone.
B6 also already carries its own EU/UK-authorised health claims, separate from this new trial evidence: normal psychological function, normal function of the nervous system, and a reduction in tiredness and fatigue [4][5]. How Does B6 Work With Magnesium?
Vitamin B6 rarely gets studied entirely on its own, because it works closely alongside magnesium in the body, and the research reflects that.

The mechanism makes sense biologically. Magnesium is involved in over 300 enzyme reactions in the body, and several of those also require B6 as a cofactor to actually function. Having enough of one nutrient without the other can limit what either can do. This is why magnesium and B6 are so often paired in the same formulation rather than sold as separate single-nutrient products.

B12, Folate and the Homocysteine Connection
B6 isn’t the only B vitamin with a genuine regulatory claim behind it. Vitamin B12 has a parallel EU/UK-authorised claim for normal neurological and psychological function, alongside normal energy-yielding metabolism and a reduction in tiredness and fatigue [7]. Folate carries its own authorised claim for normal psychological function and reduction of tiredness and fatigue [8]. All three vitamins’ claims are based on formal EFSA scientific opinions, reviewed and approved rather than simply asserted on a label.
B12 and folate share another job together: regulating homocysteine, an amino acid the body produces as a normal by-product of metabolism. When B12 or folate levels are too low, homocysteine can build up, and raised homocysteine has been linked to nervous system and cardiovascular problems. B12 deficiency is worth taking seriously in its own right, since it’s one of the few nutrient deficiencies that can cause genuine nerve damage if left unaddressed for long enough. Vegans and vegetarians are at meaningfully higher risk, since B12 occurs almost exclusively in animal foods, as are older adults, whose ability to absorb B12 from food naturally declines with age.
How Much B6 Is Safe?
This is where the B6 story needs an honest caveat, because the doses used in the trials above are far higher than what’s considered safe for ongoing, unsupervised use. The 2026 depression trial used 100mg a day for one month. The 2022 anxiety trial used roughly 50 times the standard recommended amount, around 70mg. Both were short, closely monitored research studies, and neither reported side effects during that period.
UK guidance is more conservative for anyone taking a supplement long-term without medical supervision. The NHS advises against taking more than 10mg of vitamin B6 a day from supplements unless a doctor has told you to. It also warns that 200mg or more a day, taken over time, can cause peripheral neuropathy: a loss of feeling in the hands and feet that’s occasionally permanent [9]. The European Food Safety Authority reached a similar conclusion in 2023, lowering its own tolerable upper intake level for B6 from 25mg to 12mg a day [10].
These two pictures aren’t actually in conflict. They’re answering different questions. The trial doses show what a high, short-term, supervised dose can do for mood in a research setting. The UK and EU guidance addresses a separate question: what’s safe to take yourself, every day, indefinitely, without medical oversight. A one-month research trial monitored by clinicians and an over-the-counter supplement taken for years unsupervised carry very different risk profiles, even at similar doses.
Some manufacturers use the active form of B6, pyridoxal-5-phosphate, and market it as gentler on the nerves than standard pyridoxine. There’s a plausible mechanism behind that claim: most reported cases of B6-related nerve damage have involved standard pyridoxine rather than the active form. But there’s no head-to-head safety trial comparing the two directly, and regulators set their upper limits based on total B6 intake regardless of form. Treat it as an open question rather than a settled safety advantage.
Getting B Vitamins From Food First
Food remains the first place to look, and B vitamins are genuinely widespread across a normal UK diet. Meat, fish, eggs and dairy provide B6, B12 and several of the other B vitamins together. Wholegrains, bananas, potatoes and fortified breakfast cereals are good B6 sources for anyone eating less meat. Leafy greens, beans and fortified cereals cover folate, and B12 specifically is found almost exclusively in animal products, which is why vegans in particular are advised to use a fortified food or supplement [9].
For most people eating a varied diet, food alone covers daily B vitamin needs comfortably, well within the safe ranges covered above. Supplementation becomes more relevant in specific situations:
- A diagnosed deficiency

- Pregnancy, where needs for several B vitamins increase

- Genuinely high, sustained stress
Who Should Be Careful With B Vitamin Supplements?
- If you’re already taking a multivitamin, a separate B-complex, or several single-nutrient B vitamin supplements, add up your total B6 intake across all of them rather than assuming one product’s dose is the whole picture
- If you’re pregnant or breastfeeding, check with your GP or midwife before starting a B-complex, since some ingredients and doses need individual advice
- If you’re on medication that affects B vitamin levels, particularly metformin, PPIs, or anticonvulsants (epilepsy medications), ask your GP or pharmacist before adding a supplement, since your baseline needs may already be different
- If you notice tingling, numbness or pins and needles in your hands or feet after starting a B6 supplement, stop and speak to your GP, since this is the recognised early sign of peripheral neuropathy
- If you have a diagnosed B12 deficiency or pernicious anaemia (an autoimmune condition stopping B12 absorption), this needs proper medical management rather than a general supplement, since the underlying cause needs treating directly

Is it safe to take B vitamins and magnesium together?

Can you get too much vitamin B12 or folate, the way you can with B6?
B12 and folate have a much wider safety margin than B6. The NHS considers up to 2mg of B12 a day, and up to 1mg of folate a day, unlikely to cause harm. Folate above 1mg can mask the symptoms of a B12 deficiency though, which is one reason not to self-treat a suspected deficiency without a blood test first [9].
